Working with Proteins in Biotechnology

Protein is an important factor while testifying antibodies and genes. Chromatography is a technique used to isolate the various components of a mixture and this makes its application in analysis of biomolecules very important. It is used to separate and analyze the complex DNA sequences and other compounds, and also the concentration of the samples. There are many types of chromatography used in the study of biomolecules which range from DNA/RNA to recombinant proteins and antibodies. The protein purification process is being examined in this phase and are marked or categorized with compounds like antigens or biotins. To get pure proteins in the end, the labels are removed; the labels are just there to provide accurate separation of proteins. The appliance uses a property of biomolecules i.e. similarity for metals, hence various metals are used in the chromatography columns. Immobilized Metal Affinity Chromatography is an advanced and much advanced version of sympathy chromatography used in credentials of biomolecules in present times.

The storage of these proteins

Protecting the protein structure becomes excessively important in a storage having an environment with thermodynamics equilibrium. To understand protein stabilization in details we require understanding the thermodynamics laws that govern the structural transitions of protein and it’s interaction with the surroundings. While handling protein, we require manipulating the probability of dislocating the tertiary structures. These research and analysis processes will need an organization of protein sample preparation, purification and proper storage. Though different protein classes require different handling methods but in general the above stages apply aptly. The medical researchers maintain lot of precautions while keeping these proteins perfectly. Protein solutions should be prepared in buffers and that too at high concentrations. Both the native structure of the compound and the inhibiting sticking is stabilized by the high concentration buffer. The scientists never oxygenate the sample buffer. It is a mandatory process where the biotechnology researchers maintain hygiene by wearing gloves is wore for every researcher since protein itself can be contaminated. This is because our skin contains chemical called proteases. If proteases come in contact with protein, the latter will run the risk of getting degraded. Consequently, the protein activity will be destroyed.
